Biopharmaceutical evaluation of preparations with hydrocortisone

Summary
Introduction. The release of the drug from dosage form significantly affects its pharmacological effect. The drug release from the semi-solid dosage forms depends on the physico-chemical characteristics of the drug substance - solubility of the drug, its particle size and on the properties of ointment base and the presence of excipients. Hydrocortisone is a drug commonly used in the treatment of many skin diseases, mainly in the form of ointments and creams.
Aim. The aim of this study was to evaluate of hydrocortisone release from ointments made with using various bases (white petrolatum, hydrophilic petrolatum, Hascobase, eucerin, Lecobase lux).
Material and methods. The study of hydrocortisone release was performed using a paddle apparatus with extraction chamber. The extraction chamber with the tested preparation and dialysis membrane (cellulose film Cuprophan®) were placed in a beaker with the acceptor fluid (acetate buffer, pH 5.5 containing 1% Tween 80). The test was carried out at 32±0.5°C for 6 h. The content of hydrocortisone in acceptor fluid samples were determined by high performance liquid chromatography.
Results. The obtained results indicate that the best release profile of hydrocortisone provides the formulation P2. The release of hydrocortisone was setting slowly from preparations P1, P3 and P4. This study showed no significant difference in the amount of released hydrocortisone using the hydrophilic and lipophilic membrane.
Conclusions. In the study of pharmaceutical availability of hydrocortisone the best release profile was obtained in the formulation prepared with using Hascobase - absorbing hydrated medium.
