Interactions of DNA platinum adducts with proteins and remedial mechanisms of these adducts during therapy of malignant melanomaŁukasz Glinka, Justyn Ochocki Zakład Chemii Bionieorganicznej Katedry Chemii Medycznej Uniwersytetu Medycznego w Łodzi Summary One of the most malignant cancers is malignant melanoma which almost always attacks the skin. One of the therapeutic methods of the treatment of this tumour is combined chemoimmunotherapy using cisplatin. Bioactivity of complexes containing Pt2+ ion depends on specific products formed with DNA nitrous bases. The most important adduct appeared to be intracyclic bridges situated near bases, containing jointly up to 90% of bounded platinum. The resultant inter- and intrastrand cross-links determine significant structural changes in DNA helix molecule, which in turn inhibits replication processes and induces repair mechanisms. The basic mechanism aimed at elimination of DNA distortions induced by cis-diamminedichloroplatinum (II) is NER which consists in nucleotide excision. Key words: malignant melanoma, cis-diamminedichloroplatinum(II), DNA adducts, HMG proteins, NER pathway |
